Abstract
Unlimited proliferation due to the disruption of the cell cycle is a distinctive property in tumor cells. In recent decade, MCM proteins have been much attractive as new target molecules in the check point pathway of the cell cycle progression. MCM proteins are essential for eukaryotic DNA replication and its expression is much higher in malignant tissues than normal tissues. Therefore, the agents that down-regulate MCM proteins expression might contribute to cancer prevention or therapy. The screening of up to 200 extracts from natural medicinal plants led to the discovery of Ficus carica as candidate. Ficus carica leaves were extracted with methanol and then fractionated with CH2Cl2, EtOAC, n-BuOH and H2O. The n-BuOH fraction showed the down-regulation activity of MCM protein expression. The n-BuOH fraction was further purified to isolate active substance by column chromatography. Isoschaftoside (Apigenin 6-C-¥á-L-Arabinopyranosyl-8-C-¥â-D-glucopyranoside) was isolated and identified from n-BuOH fraction as a major active component of MCM protein expression regulator. The findings of our present study merit further research on the application of isoschaftoside as a potential agent for treatment of cancer.
